Quantitative proteomics analysis of Mycoplasma pneumoniae identifies potential macrolide resistance determinants

چکیده

Abstract Mycoplasma pneumoniae is one of the leading causes community-acquired pneumonia in children and adolescents. Because wide application macrolides clinical treatment, macrolide-resistant M. strains have become increasingly common worldwide. However, molecular mechanisms underlying drug resistance are poorly understood. In present work, we analyzed whole proteomes macrolide-sensitive using a tandem mass tag-labeling quantitative proteomic technique, Data available via ProteomeXchange with identifier PXD022220. total, 165 differentially expressed proteins were identified, which 80 upregulated 85 downregulated drug-resistant strain compared sensitive strain. Functional analysis revealed that these predominantly involved protein peptide biosynthesis processes, ribosome, transmembrane transporter activity, implicates them mechanism(s) to macrolides. Our results provide new insights into identify potential targets for further studies on this bacterium.

Macrolide Resistance in Mycoplasma pneumoniae, Israel, 2010

Macrolide resistance in Mycoplasma pneumoniae is often found in Asia but is rare elsewhere. We report the emergence of macrolide-resistant M. pneumoniae in Israel and the in vivo evolution of such resistance during the treatment of a 6-year-old boy with pneumonia.

Macrolide Resistance of Mycoplasma pneumoniae, South Korea, 2000–2011

In Korea, Mycoplasma pneumoniae was detected in 255/2,089 respiratory specimens collected during 2000-2011; 80 isolates carried 23S rRNA gene mutations, and 69/123 culture-positive samples with the mutation were resistant to 5 macrolides. During 2000-2011, prevalence of the mutation increased substantially. These findings have critical implications for the treatment of children with mycoplasma ...

Drug Resistance Mechanisms of Mycoplasma pneumoniae to Macrolide Antibiotics

Throat swabs from children with suspected Mycoplasma pneumoniae (M. pneumoniae) infection were cultured for the presence of M. pneumoniae and its species specificity using the 16S rRNA gene. Seventy-six M. pneumoniae strains isolated from 580 swabs showed that 70 were erythromycin resistant with minimum inhibitory concentrations (MIC) around 32-512 mg/L. Fifty M. pneumoniae strains (46 resistan...

Mycoplasma pneumoniae: Current Knowledge on Macrolide Resistance and Treatment

Mycoplasma pneumoniae causes community-acquired respiratory tract infections, particularly in school-aged children and young adults. These infections occur both endemically and epidemically worldwide. M. pneumoniae lacks cell wall and is subsequently resistant to beta-lactams and to all antimicrobials targeting the cell wall. This mycoplasma is intrinsically susceptible to macrolides and relate...
